    A new method for detecting differential item functioning in the Rasch model

    Differential item functioning (DIF) can lead to an unfair advantage or disadvantage for certain subgroups in educational and psychological testing. Therefore, a variety of statistical methods has been suggested for detecting DIF in the Rasch model. Most of these methods are designed for the comparison of pre-specified focal and reference groups, such as males and females. Latent class approaches, on the other hand, allow to detect previously unknown groups exhibiting DIF. However, this approach provides no straightforward interpretation of the groups with respect to person characteristics. Here we propose a new method for DIF detection based on model-based recursive partitioning that can be considered as a compromise between those two extremes. With this approach it is possible to detect groups of subjects exhibiting DIF, which are not prespecified, but result from combinations of observed covariates. These groups are directly interpretable and can thus help understand the psychological sources of DIF. The statistical background and construction of the new method is first introduced by means of an instructive example, and then applied to data from a general knowledge quiz and a teaching evaluation

    Anchor methods for DIF detection: A comparison of the iterative forward, backward, constant and all-other anchor class

    In the analysis of differential item functioning (DIF) using item response theory (IRT), a common metric is necessary to compare item parameters between groups of test-takers. In the Rasch model, the same restriction is placed on the item parameters in each group in order to define a common metric. However, the question how the items in the restriction - termed anchor items - are selected appropriately is still a major challenge. This article proposes a conceptual framework for categorizing anchor methods: The anchor class to describe characteristics of the anchor methods and the anchor selection strategy to guide how the anchor items are determined. Furthermore, a new anchor class termed the iterative forward anchor class is proposed. Several anchor classes are implemented with two different anchor selection strategies (the all-other and the single-anchor selection strategy) and are compared in an extensive simulation study. The results show that the newly proposed anchor class combined with the single-anchor selection strategy is superior in situations where no prior knowledge about the direction of DIF is available. Moreover, it is shown that the proportion of DIF items in the anchor - rather than the fact whether the anchor includes DIF items at all (termed contamination in previous studies) - is crucial for suitable DIF analysis

    Anchor selection strategies for DIF analysis: Review, assessment, and new approaches

    Differential item functioning (DIF) indicates the violation of the invariance assumption for instance in models based on item response theory (IRT). For item-wise DIF analysis using IRT, a common metric for the item parameters of the groups that are to be compared (e.g. for the reference and the focal group) is necessary. In the Rasch model, therefore, the same linear restriction is imposed in both groups. Items in the restriction are termed the anchor items. Ideally, these items are DIF-free to avoid artificially augmented false alarm rates. However, the question how DIF-free anchor items are selected appropriately is still a major challenge. Furthermore, various authors point out the lack of new anchor selection strategies and the lack of a comprehensive study especially for dichotomous IRT models. This article reviews existing anchor selection strategies that do not require any knowledge prior to DIF analysis, offers a straightforward notation and proposes three new anchor selection strategies. An extensive simulation study is conducted to compare the performance of the anchor selection strategies. The results show that an appropriate anchor selection is crucial for suitable item-wise DIF analysis. The newly suggested anchor selection strategies outperform the existing strategies and can reliably locate a suitable anchor when the sample sizes are large enough

    In 1908, ten iron military equipment items were discovered in Brigantium/Bregenz (Austria). Among them are fragments of four or five shield bosses of rectangular scuta. This is the largest ensemble of this archaeologically rare type of umbo hitherto known. They are probably preserved because they were burnt and therefore the material could not be recycled. The typological dating of the shield bosses indicates that they were buried during the dismantling of the fort in Brigantium in the early Claudian Period or shortly before. It remains unclear in which constructional context the objects were recovered, because the stratigraphic circumstances of finding were not documented and there is not enough knowledge about the military buildings on the excavation area. Due to the location within the fort, it seems most likely that it was a barrack or a magazine building situated along the via principalis.En 1908 se encontraron diez elementos de equipamiento militar de hierro en Brigantium/Bregenz (Austria). Entre estos figuran los fragmentos de cuatro o cinco umbos de scuta rectangulares. Se trata del conjunto de este tipo de umbos, arqueológicamente insólitos, más grande conocido. Probablemente su buen estado de conservación se debe a que las piezas estaban quemadas y su reciclado resultaba imposible. La datación tipológica de los umbos indica que fueron enterrados durante del cierre de la fortaleza en Brigantium en la época claudiana o poco antes. No está claro en qué contexto estructural fueron recuperados los objetos, porque las condiciones estratigráficas no fueron documentadas e y no se sabe lo suficiente acerca de los edificios militares en el área de la excavación. Debido a su situación dentro de la fortaleza, parece probable que se tratase de un barracón o almacén localizado en la via principalis

    Model-based recursive partitioning meets item response theory

    The aim of this thesis is to develop new statistical methods for the evaluation of assumptions that are crucial for reliably assessing group-differences in complex studies in the field of psychological and educational testing. The framework of item response theory (IRT) includes a variety of psychometric models for scaling latent traits such as the widely-used Rasch model. The Rasch model ensures objective measures and fair comparisons between groups of subjects. However, this important property holds only if the underlying assumptions are met. One essential assumption is the invariance property. Its violation is extensively discussed in the literature and termed differential item functioning (DIF). This thesis focuses on the methodology of DIF detection. Existing methods for DIF detection are briefly discussed and new statistical methods for DIF detection are introduced together with new anchor methods. The methods introduced in this thesis allow to classify items with and without DIF more accurately and, thus, to improve the evaluation of the invariance assumption in the Rasch model. This thesis, thereby, provides a contribution to the construction of objective and fair tests in psychology and educational testing

    Epigenetic Regulation of S100A9 and S100A12 Expression in Monocyte-Macrophage System in Hyperglycemic Conditions

    The number of diabetic patients in Europe and world-wide is growing. Diabetes confers a 2-fold higher risk for vascular disease. Lack of insulin production (Type 1 diabetes, T1D) or lack of insulin responsiveness (Type 2 diabetes, T2D) causes systemic metabolic changes such as hyperglycemia (HG) which contribute to the pathology of diabetes. Monocytes and macrophages are key innate immune cells that control inflammatory reactions associated with diabetic vascular complications. Inflammatory programming of macrophages is regulated and maintained by epigenetic mechanisms, in particular histone modifications. The aim of our study was to identify the epigenetic mechanisms involved in the hyperglycemia-mediated macrophage activation. Using Affymetrix microarray profiling and RT-qPCR we identified that hyperglycemia increased the expression of S100A9 and S100A12 in primary human macrophages. Expression of S100A12 was sustained after glucose levels were normalized. Glucose augmented the response of macrophages to Toll-like receptor (TLR)-ligands Palmatic acid (PA) and Lipopolysaccharide (LPS) i.e., pro-inflammatory stimulation. The abundance of activating histone Histone 3 Lysine 4 methylation marks (H3K4me1, H3K4me3) and general acetylation on histone 3 (AceH3) with the promoters of these genes was analyzed by chromatin immunoprecipitation. Hyperglycemia increased acetylation of histones bound to the promoters of S100A9 and S100A12 in M1 macrophages. In contrast, hyperglycemia caused a reduction in total H3 which correlated with the increased expression of both S100 genes. The inhibition of histone methyltransferases SET domain-containing protein (SET)7/9 and SET and MYND domain-containing protein (SMYD)3 showed that these specifically regulated S100A12 expression. We conclude that hyperglycemia upregulates expression of S100A9, S100A12 via epigenetic regulation and induces an activating histone code on the respective gene promoters in M1 macrophages. Mechanistically, this regulation relies on action of histone methyltransferases SMYD3 and SET7/9. The results define an important role for epigenetic regulation in macrophage mediated inflammation in diabetic conditions

    Metabolic Activation of Intrahepatic CD8+ T Cells and NKT Cells Causes Nonalcoholic Steatohepatitis and Liver Cancer via Cross-Talk with Hepatocytes

    SummaryHepatocellular carcinoma (HCC), the fastest rising cancer in the United States and increasing in Europe, often occurs with nonalcoholic steatohepatitis (NASH). Mechanisms underlying NASH and NASH-induced HCC are largely unknown. We developed a mouse model recapitulating key features of human metabolic syndrome, NASH, and HCC by long-term feeding of a choline-deficient high-fat diet. This induced activated intrahepatic CD8+ T cells, NKT cells, and inflammatory cytokines, similar to NASH patients. CD8+ T cells and NKT cells but not myeloid cells promote NASH and HCC through interactions with hepatocytes. NKT cells primarily cause steatosis via secreted LIGHT, while CD8+ and NKT cells cooperatively induce liver damage. Hepatocellular LTβR and canonical NF-κB signaling facilitate NASH-to-HCC transition, demonstrating that distinct molecular mechanisms determine NASH and HCC development

    The ocean sampling day consortium

    Ocean Sampling Day was initiated by the EU-funded Micro B3 (Marine Microbial Biodiversity, Bioinformatics, Biotechnology) project to obtain a snapshot of the marine microbial biodiversity and function of the world’s oceans. It is a simultaneous global mega-sequencing campaign aiming to generate the largest standardized microbial data set in a single day. This will be achievable only through the coordinated efforts of an Ocean Sampling Day Consortium, supportive partnerships and networks between sites. This commentary outlines the establishment, function and aims of the Consortium and describes our vision for a sustainable study of marine microbial communities and their embedded functional traits
